Welcome back to New Directions and Approaches in the Management of Patients with HER2-Altered Non-Small Cell Lung Cancer. This is medical oncologist Dr Neil Love. I met with Dr Joshua Sabari from the NYU Langone Health Perlmutter Cancer Center in New York City to discuss key presentations and publications on one of the most exciting new areas in oncology research,

particularly the recent approval of two HER2-directed tyrosine kinase inhibitors, Zongertinib and Sevabirtinib. In the first part of this series, Dr Sabari discussed three cases. In the next two parts, he reviews a number of key presentations and publications, including perhaps the Non-Small Cell Lung Cancer Paper of the Year from the New England Journal of Medicine,

Zongertinib and previously treated HER2-mutant non-small cell lung cancer. John Hamak from MD Anderson presented this at AACR 2025. It was Zongertinib and previously treated HER2-mutant metastatic non-small cell lung cancer. Remember, these are patients that have had prior therapy, so this is not frontline here. And what we saw up front was a robust response rate, 71% of patients in response.

You also saw a very meaningful progression-free survival here at 12.4 months, and it was well tolerated. So for me, I agree with you, Neil. I think this was one of the first representations of a truly targeted agent in the HER2 space. This was practice changing. This study led to an accelerated FDA approval in the second-line setting. Obviously, there is a confirmatory Phase III study ongoing right now.

You know, when this data came out, you know, I really saw a huge transition in patients, you know, or in clinicians' focus on how to treat this mutation. A lot of patients are already getting or were already getting trastuzumab, duroxatecan in clinical setting in the second line.
