Hi, I'm John Heymach. I'm the Chair of Thoracic Head and Neck Medical Oncology at MD Anderson. And I'm thrilled to be presenting here about all the exciting progress over the past year in targeting HER2, MET, and KRAS G12C. Really an exciting year of progress.

So let me start off with HER2-altered non-small cell lung cancer. And this has really been a rapidly evolving landscape. Let's start off by reminding everybody here I'm talking about HER2 mutants. In lung cancer, HER2 mutants are the thing that we are focused on treating for the most part.

HER2 amplifications and overexpression are another group, but they're not one that we are typically focused on in our day-to-day treatment. HER2 mutants are really bonafide oncogenic drivers.

HER2 mutants are really bonafide oncogenic drivers. And, you know, for the last several years, our treatment paradigm has been starting with platinum doublet with or without immunotherapy, followed by tristuzumab-duruxtacin, which has had FDA approval in the post-platinum setting. And then after that, our treatment options are limited to dosetaxin and ramissurumab. Now, last year, we had the approval of two drugs in the second-line setting.

That's zungertinib, an oral tyrosine kinase inhibitor, and sevoburtinib, an additional TKI. So those became options then in the post-platinum setting. And then after progression on one or the other, again, with three different second-line options, then we had decisions about which one to switch to next and then dosetaxin remaining an option.

Well, now we've recently had first-line approval of zungertinib for HER2 mutant non-small cells. So that's really shifted the entire landscape. So now if you start with an oral tyrosine kinase inhibitor, we have three different options. You can go to the antibody drug conjugate, trastuzumab-druxtacin. You can go to platinum doublet or sevoburtinib.
