New leadership and new partnerships have helped Pfizer revitalise its oncology pipeline, and a strong showing at the recent American Society of Clinical Oncology annual meeting for some of its novel programmes, plus a clear focus and prioritisation for its cancer assets,

have the company poised for growth, Chief Oncology Officer Jeff Lagos told Scrips Mary Jo Laffler. More than half of Pfizer's Phase III cancer programmes are for established products, 13 versus 6 for novel compounds. But those numbers reverse earlier in the pipeline, with 5 product enhancements to 4 novel compounds in Phase II, and 5 product enhancements to 19 novel programmes in Phase I.

Pfizer has also been signing collaborations, including with China's Innovent Biologics for early-stage antibody drug conjugate and multi-specific antibody programmes to build its earlier oncology pipeline. Oncology is one of Pfizer's priority therapeutic areas, along with cardiometabolic slash primary care, inflammation, immunology and vaccines, and Lagos was brought on board last year as Chief Oncology Officer after running Novartis Oncology for a decade.

Pfizer is aligning its research into a few core areas, including antibody drug conjugates and multi-specific antibodies, Lagos explained in an interview with Scrip. The therapeutic focus is on breast, geniturinary, gastrointestinal and lung cancers, plus haematology. If we have an approved medicine where we understand the science, and it has already provided significant

impact to patients, we are trying to move that into earlier lines of therapy, earlier stages of disease, he explained. In lung cancer, for example, Pfizer has multiple Phase III trials underway for its PD-1 times VEGF targeting, bispecific antibody PF4404, in collaboration with Chinese biopharma 3S-Bio,

and the integrin beta-6ADC-sigvotatug vedetin, inherited from Seagun. Early data for both were released at ASCO. Pfizer is also awaiting Phase III data for its EZH2 inhibitor, mevromatostat, in first-line metastatic castration-resistant, as well as castration-sensitive prostate cancer.
