That's how American Society of Clinical Oncology Chief Medical Officer Julie Gralow Scrip described results from the Phase 3 RASOLOOT-302 trial of Revolution Medicine's daraxonrasib in previously treated metastatic pancreatic adenocarcinoma during a press briefing at the 2026 ASCO meeting, Alaric de Armond writes. The US biotech presented data from the 500-patient study of its RAS/ONT inhibitor during the meeting in Chicago, and the company told Scrip it is actively preparing global regulatory submissions, including in the US, which will incorporate the new data. During the May 30 press briefing, presenter Brian Wolpin, director of the Gastrointestinal Cancer Center at the Dana-Farber Cancer Institute, highlighted the inadequacy of current chemotherapy options for pancreatic cancer, where RAS signaling is an important driver of tumor growth. More than 90% of PDAC tumors harbor an oncogenic RAS mutation, but there have been no approved therapies that target RAS that can be used for pancreatic cancer. In the RASALUTE trial, RAS G12 mutation-positive population, median overall survival was 13.2 months for patients receiving daraxonrasib versus 6.6 months for patients in the control arm, which received investigator's choice of chemotherapy. Median OS was similar in the overall population, —with a 13.2-month OS for patients in the daraxonrasib arm and 6.7 months in the control arm. Progression-free survival by blinded independent central review was 7.3 months in the RAS G12 mutation-positive population versus 3.5 months in the control arm. In the overall population, those figures were 7.2 months and 3.6 months respectively. The discussant, University of Arizona Cancer Center oncologist Rashna Shroff, called the results a "game changer," noting in particular the doubling in OS and improvements in PFS versus chemotherapy. At the same time, she noted that there have been questions around daraxonrasib's safety profile. Nearly 98% of patients in the daraxonrasib arm experienced treatment-related adverse events,, which led to dose reductions among 36.1% of patients receiving the drug, although Shroff said the safety profile is relatively predictable and really did not lead to a significant amount of discontinuation.
