Subsequent to the recording of this program, the FDA granted approval to Palvociclib in combination with Trastuzumab, with or without Pertuzumab and Endocrine Therapy for maintenance treatment of adults with HR-positive HER2-positive locally advanced or metastatic breast cancer following induction treatment based on results of the Phase III PATINA trial.

So, hello everybody. My name is Sibylle Loibl. I'm from the University Hospital in Frankfurt and the German Breast Group as well in Frankfurt. I'm a trained gynecologist and I'm doing breast cancer and breast cancer research since many, many years and I hope we have a good interaction today. So, I will be speaking about the first-line and maintenance therapy for patients for HER2-positive metastatic breast cancer.

So, I think before we go into the detail of the first-line metastatic breast cancer patients, I would like to make you aware of the recent changes we have seen in metastatic breast cancer in the HER2-positive field in general.

You can see here the outcome in the metastatic breast cancer patients starting with the third-line where TDXT based on the DESTINY BREAST-02 study was actually compared with capecitabine, trastuzumab or capecitabine, lapatinib. And you can see here that the median PFS was significantly longer from 7 to 17.8 months.

This also translated overall in an overall survival benefit associated with TDXT. The next study which was almost running in parallel was the DBO3 study which compared actually two ADCs, TDXT and TDM1 which was the standard of care at that time.

And as you can see here, again, the median progression-free survival was significantly improved from 7 to 29 months. And the overall survival, again, could be improved with such a large difference.
